Thioredoxin Reductase Inhibition as a Novel Therapy in Cystic Fibrosis

We have previously shown that inhibition of the redox enzyme thioredoxin reductase (TrxR) strongly induces endogenous antioxidant responses that are protective in models of oxidant lung injury. Coincident with enhanced antioxidant expression is increased production of a major intracellular antioxidant, glutathione (GSH). Cystic Fibrosis (CF) is a chronic lung disease with high morbidity and mortality and GSH levels are known to be low in CF. These preclinical studies will investigate the effects of TrxR inhibition on epithelial physiology in cell and animal models of CF with particular emphasis on GSH production.
